    A physicochemical perspective of aging from single-cell analysis of pH, macromolecular and organellar crowding in yeast

    Cellular aging is a multifactorial process that is characterized by a decline in homeostatic capacity, best described at the molecular level. Physicochemical properties such as pH and macromolecular crowding are essential to all molecular processes in cells and require maintenance. Whether a drift in physicochemical properties contributes to the overall decline of homeostasis in aging is not known. Here we show that the cytosol of yeast cells acidifies modestly in early aging and sharply after senescence. Using a macromolecular crowding sensor optimized for long-term FRET measurements, we show that crowding is rather stable and that the stability of crowding is a stronger predictor for lifespan than the absolute crowding levels. Additionally, in aged cells we observe drastic changes in organellar volume, leading to crowding on the µm scale, which we term organellar crowding. Our measurements provide an initial framework of physicochemical parameters of replicatively aged yeast cells

    Influence of Snowmelt Timing on the Diet Quality of Pyrenean Rock Ptarmigan (Lagopus muta pyrenaica): Implications for Reproductive Success

    The Pyrenean rock ptarmigan (Lagopus muta pyrenaica) is the southernmost subspecies of the species in Europe and is considered threatened as a consequence of changes in landscape, human pressure, climate change, and low genetic diversity. Previous studies have shown a relationship between the date of snowmelt and reproductive success in the Pyrenean ptarmigan. It is well established that birds laying early in the breeding season have higher reproductive success, but the specific mechanism for this relationship is debated. We present an explicative model of the relationship between snowmelt date and breeding success mediated by food quality for grouse in alpine environments. From microhistological analyses of 121 faecal samples collected during three years in the Canigou Massif (Eastern Pyrenees), and the assessment of the chemical composition of the main dietary components, we estimated the potential quality of individual diets. Potential dietary quality was correlated with free-urate faecal N, a proxy of the digestible protein content ingested by ptarmigan, and both were correlated with phenological stage of consumed plants, which in turn depends on snowmelt date. Our findings suggest that the average snowmelt date is subject to a strong interannual variability influencing laying date. In years of early snowmelt, hens benefit from a longer period of high quality food resources potentially leading to a higher breeding success. On the contrary, in years of late snowmelt, hens begin their breeding period in poorer nutrient condition because the peaks of protein content of their main food items are delayed with respect to laying date, hence reducing breeding performance.     Managers' Corporate Social Responsibility Perceptions and Attitudes across Different Organizational Contexts within the Non-Profit-For-Profit Organizational Continuum

    This article presents qualitative, case study research and contributes to the literature by presenting managers' views on corporate social responsibility (CSR) within three different types of organizational contexts (non-profit, for-profit and a middle/hybrid case) that have never embedded CSR policies and practices. To the best of the author's knowledge, no other study has compared managers' CSR attitudes within such a combination of organizational contexts. The findings from this study indicate that some considerations with regards to CSR are transferable across organizational contexts (i.e. issues around the attainability of CSR, emphasis given to ethics and integrity and managers associating CSR with employee-related activities), while others are context specific. The context-specific ones show that organizational culture and an organization's position in the non-profit/for-profit continuum are key contextual influences on managers' CSR attitudes. Moreover, influences relating to individual managers' work, as well as the extent to which CSR is an implicit or an explicit part of decision-making determine managerial perceptions of CSR. The implications for management practice (i.e. a one-size-fits-all approach to CSR practice is wrong) and CSR research (i.e. more context-sensitive and interdisciplinary research) are discussed. The findings are relevant both to organizations considering CSR engagement and those already implementing CSR. © 2012 Copyright Taylor and Francis Group, LLC

    Poor replication of candidate genes for major depressive disorder using genome-wide association data.

    Data from the Genetic Association Information Network (GAIN) genome-wide association study (GWAS) in major depressive disorder (MDD) were used to explore previously reported candidate gene and single-nucleotide polymorphism (SNP) associations in MDD. A systematic literature search of candidate genes associated with MDD in case-control studies was performed before the results of the GAIN MDD study became available. Measured and imputed candidate SNPs and genes were tested in the GAIN MDD study encompassing 1738 cases and 1802 controls. Imputation was used to increase the number of SNPs from the GWAS and to improve coverage of SNPs in the candidate genes selected. Tests were carried out for individual SNPs and the entire gene using different statistical approaches, with permutation analysis as the final arbiter. In all, 78 papers reporting on 57 genes were identified, from which 92 SNPs could be mapped. In the GAIN MDD study, two SNPs were associated with MDD: C5orf20 (rs12520799; P0.038; odds ratio (OR) AT1.10, 95% CI 0.95-1.29; OR TT1.21, 95% confidence interval (CI) 1.01-1.47) and NPY (rs16139; P=0.034; OR C allele0.73, 95% CI 0.55-0.97), constituting a direct replication of previously identified SNPs. At the gene level, TNF (rs76917; OR T1.35, 95% CI 1.13-1.63; P0.0034) was identified as the only gene for which the association with MDD remained significant after correction for multiple testing. For SLC6A2 (norepinephrine transporter (NET)) significantly more SNPs (19 out of 100; P0.039) than expected were associated while accounting for the linkage disequilibrium (LD) structure. Thus, we found support for involvement in MDD for only four genes. However, given the number of candidate SNPs and genes that were tested, even these significant may well be false positives. The poor replication may point to publication bias and false-positive findings in previous candidate gene studies, and may also be related to heterogeneity of the MDD phenotype as well as contextual genetic or environmental factors. © 2011 Macmillan Publishers Limited All rights reserved
